How can I tell the difference between a m20 and m21 gearbox. Also how will I know it is for a 253 or 308

Assuming you mean aussie 4spd, put them in 1st gear and cound how many turns the input shaft does for 1 revolution of the output shaft. 3.05 for M20, 2.54 for M21. There is no difference between 253 and 308 boxes (apart from only M21 being available for 3

circular grooves on input shaft
1 for m 20
2 for m21
exception is XU1 m21 no grooves, is called m21 but has diff ratios

Not quite. The XU-1 box (899 cluster) is called an XU-1 M20. There is also an optional XU-1 box with the same ratios as the V8 M21, and another with the L34 ratios. You also can't rely upon the grooves due to aftermarket stuff with wrong grooves (or none)
