Good evening.If I remove the tail shaft block up the transmission to hold it up then remove the transmission mount is there any reason or anything
to watch out for in removing the extension housing from a Trimatic in a HJ Holden whilst in position?

Transmission fluid. I’d suggest you have a new gasket and rear seal on hand and change them whilst the end housing is off. Put some fluid on the new seal before putting the yoke back in. Also best get the rubber seals for the speedo cable end.
