Does any know if the bore size of a Holden V8 engine can be sustanciated between 308 vs 253 without pulling off the cylinder head?

Just imagine there is no engine number and the displacement no on the side of the block has been ground-off.

No. The stock bore size and corresponding components are different one to the other. The 308 has a 4" bore and the 253 is 3.625".

253 is 3.625
same as 202 holden

I think what SuSA9X wants to know is, how does he tell a 253 from a 308, without removing the head, if the engine number and casting number are not there.

The motors are the same externally. However, an early 253 had a 2 barrel carb and its manual box was usually an M20 rather than an M21, the Trimatic and diff were the same, except in the HK and HG where the 308 had a Salisbury.

Is there a cylinder volume test (i.e. Air or fluid) that can be done to measure displacement or are there collapsible Verniers available which can be inserted through the spark plug hole?

I would have thought, given that these things have been around s

CAMS scrutineers would probably have a cc tester (or more accurately a cylinder volume tester). Ive never seen one though.
